ThispremadelentiviralparticlesisthesinglevectormethodforiPScellgeneration.FourhumaniPSfactor,c-Myc,Klf4,Oct4andSox2(MKOS)wereclonedunderthesameTetracyclineinducIBLeCMVpromoter.All4factorswereclonedinframebutseparatedby2Asignalfragment,whichresultintheexpressionoffourindividualfactorvia“ribosomalskipping”mechanism.Inaddition,theparticlescontainsaGFP-blasticidindualselectionMarkerunderRSVpromoter,whichprovidesaconvenience methodtomonitortheiPSfactorexpressionefficiency,andtosortthepositivetransduced cells.
ThefouriPSfactorareexpressedunderanoptionalinducibleCMVpromoter.ThepremadeparticlecanbeusedasconstitutiveexpressionoffouriPSfactors,butoptionally,itcanbeusedasantetracyclineinducibleexpressionofiPSfactors.Tousedasinducibleexpression,therepressprotein(TetR)mustbepresentinadvancetostoptheexpression,theexpressionofiPSfactorsonlyoccurswhentheTetracyclineorDoxwasaddedintothecellculture.PleasetoseemoredetailsabouttheOptionalinduciblelentiviralexpression.
